M\u00e9xico.-\u00a0 <strong>En dos modelos in vitro, la doctora en Ciencias Bioqu\u00edmicas, Magdalena Guerra Crespo, y su alumno de maestr\u00eda, Francisco Javier Padilla God\u00ednez, probaron un dispositivo nanotecnol\u00f3gico para hacer llegar directamente la dopamina al cerebro, a fin de garantizar su difusi\u00f3n constante en pacientes con la enfermedad de Parkinson.<\/strong><\/p>\n<p><strong>Guerra Crespo, investigadora del Departamento de Fisiolog\u00eda de la Facultad de Medicina (FM) de la UNAM, detall\u00f3 que este dispositivo tiene una matriz amorfa (estructura peque\u00f1a sin una forma precisa) y porosa, menor a 100 nan\u00f3metros.<\/p>\n<p>\u201cLa dopamina no se puede administrar a los pacientes como tal porque se oxida, por eso se utiliza su precursor, la levodopa, que se receta en tabletas v\u00eda oral\u201d, precis\u00f3 la cient\u00edfica quien estudia el padecimiento hace m\u00e1s de dos d\u00e9cadas.<\/p>\n<p>En el estudio en el cual tambi\u00e9n colabor\u00f3 Tessy Mar\u00eda L\u00f3pez Goerne, acad\u00e9mica de la Universidad Aut\u00f3noma Metropolitana Xochimilco (UAM-X), Padilla God\u00ednez desarroll\u00f3 el proyecto en laboratorios de la FM de la UNAM y de la UAM; los resultados fueron publicados recientemente en la revista internacional\u00a0<em>Nanomedicine.<\/em><\/p>\n<p>Estas dimensiones a nanoescala se pueden imaginar cuando sabemos que un nan\u00f3metro es equivale a la milmillon\u00e9sima parte del metro; un cabello humano tiene aproximadamente 60 mil nan\u00f3metros de espesor, precis\u00f3.<\/p>\n<p>\u201cEsta matriz tiene mesoporos, es decir, poros con un tama\u00f1o entre dos y 50 nan\u00f3metros. La maravilla de esto es que el \u00e1rea superficial de esos mesoporos es de alrededor de 300 metros cuadrados por gramo\u201d. Es decir, es una superficie inmensa. Pueden contener bastante dopamina en un material con demasiados poros, como una esponja, se\u00f1al\u00f3.<\/p>\n<p>La cient\u00edfica abund\u00f3 que a esa estructura porosa se adhiere la dopamina a trav\u00e9s de interacciones qu\u00edmicas de fuentes de hidr\u00f3geno. \u201cA pesar de que los mesoporos son tan peque\u00f1itos, su superficie es muy grande, alrededor de 300 metros cuadrados por gramo\u201d.<\/p>\n<p>Actualmente la investigaci\u00f3n est\u00e1 en su fase inicial\u00a0<em>in vitro,<\/em>\u00a0y observaron que es biocompatible. Posteriormente tendr\u00e1 que probarse en un modelo animal de ratas o ratones, y a m\u00e1s largo plazo en protocolos en humanos.<\/p>\n<p>\u201cAhora vamos en el nivel experimental. Lo ideal en un modelo animal ser\u00eda realizar una cirug\u00eda estereot\u00e1xica (permite inyectar el nanoreservorio a trav\u00e9s de coordenadas) en el estriado, un \u00e1rea espec\u00edfica del cerebro donde se pierde la dopamina\u201d, coment\u00f3.<\/p>\n<p>Debido a que se necesita una cirug\u00eda para introducir el objeto nanotecnol\u00f3gico, el planteamiento de la cient\u00edfica es que se vayan creando materiales capaces de cruzar la barrera hematoencef\u00e1lica.<\/p>\n<p>Los artefactos se desarrollaron mediante colaboraci\u00f3n entre la FM de la UNAM y el laboratorio a cargo de L\u00f3pez Goerne en la UAM-X. \u201cLos sintetiz\u00f3 Francisco Javier Padilla, un estudiante muy brillante que logra su grado de maestr\u00eda con este proyecto\u201d, record\u00f3.<\/p>\n<p>Guerra Crespo expuso que se puede controlar el tama\u00f1o del poro para que la dopamina se difunda con cierta velocidad. \u201cEl tama\u00f1o del poro condiciona ese flujo, el ritmo de la dosificaci\u00f3n\u201d.<\/p>\n<p>La estructura f\u00edsico-qu\u00edmica del dispositivo nanotecnol\u00f3gico est\u00e1 unida por puentes de hidr\u00f3geno, as\u00ed la dopamina se conserva dentro del poro, queda atrapada en \u00e9l, y ya despu\u00e9s es gradualmente liberada.<\/p>\n<p>M\u00e1s adelante se podr\u00eda fabricar a medida de cada paciente para garantizar la dosificaci\u00f3n deseada; la meta ser\u00eda alargar su calidad de vida.<\/p>\n<p>\u201cTodav\u00eda faltan varios a\u00f1os, estamos en la etapa precl\u00ednica y estos an\u00e1lisis farmacol\u00f3gicos son largos.
